摘要
本实验根据马蹄铁鲎mtDNA 12SrRNA基因序列进行了中华绒螯蟹mtDNA 12SrRNA基因的引物设计,并对长江、辽河、瓯江三水系各5个个体共15个个体进行了PCR扩增及其序列测定。结果表明,12SrRNA基因序列在长江种群、辽河种群、瓯江种群间几乎没有差异,仅在3号瓯江蟹415bp处发现有一碱基颠换,去掉两端引物共测定得到415bp的碱基序列、其中A、T、G、C含量分别为145bp(34.9%),165bp(39.8%),45bp(10.8%)和60bp(14.5%)。
The primers of Eriocheir sinensis from the Changjiang, Liaohe and Oujiang Rivers were designed
according to the sequences of mitochondrial DNA 12SrRNA region of Limulus polyphemus. Besides, 415 base pairs
of encoded 12SrRNA region of mitochondrial DNA from 15 individuals were amplified and sequenced and the
contents of A, T, G and C are 145bp (34.9% ), 165bp (39.8%), 45bp (10.8% ) and 60bp (14.5%),
respectively. Among the fifteen individuals, there was only one different site in sequences, which was transversion
site at 415 base pairs of the third from the Oujiang River.
