蔗茅(Erianthus fulvus)的特异性状及其与甘蔗杂交F1代的染色体和RAPD鉴定研究 被引量:12

摘要 本文以我们的前期研究为基础 ,综述蔗茅 (Erianthusfulvus)野生种的一些特异性状及其在甘蔗育种中的利用价值。对蔗茅花粉进行低温贮存 ,克服其与甘蔗花期不遇的障碍 ,获得两个杂交组合共 310株实生苗。采用染色体计数和RAPD分子标记的方法鉴定杂种F1代真实性。结果表明 ,供试材料杂种0 1/4 7、 0 1/85、 0 1/12 0的 2n =79- 82 ,染色体遗传方式为“n +2n” ;用 110个随机引物进行RAPD扩增 ,6 3个引物可获得双亲的RAPD多态性 ,其中 3个引物 (OPC - 19、OPE - 2、OPF - 4)可在杂种0 1/6 4和 0 1/12 0的RAPD扩增标记中显示出双亲的特征谱带 ,分别为父本 35 0 0bp和母本 130 0bp、父本135 0bp和母本 90 0bp、父本 5 5 0bp和母本 90 0bp。本研究探索杂种实生苗早期鉴定的可行之法 ,为分子标记辅助选择 (MAS)、缩短甘蔗育种年限奠定工作基础。 Depended on our earlier stage research results, this paper summaried some special characteristics of Erianthus fulvus and its using value in sugarcane breeding. 310 seedlings were got by stored pollen of E.fulvus in low temperature be pollinated with sugarcane. These hybrid were identified by chromosome number and RAPD technique.The research results reveal that hybrid 01/47,01/85,and 01/120 have 2n=79-82 chromosome number, the chromosome transmission of their parents is a type of"n+2n". Of 110 RAPD primers used, 63 were polymorphic. The three primers of OPC-19, OPE-2 and OPF-4 were shown to be able to determine hybrid 01/64 and 01/120, which they possess maternal-specific and paternal-specific DNA bands, that is, 1300bp and 3500bp, 900bp and 1350bp, 900bp and 550bp, respectively.
