摘要
从两个番茄杂种L402、L404及它们的亲本共6份材料的叶片中提取DNA,使用国产PCR仪,建立了适合番茄RAPD分析的PCR条件,进一步在100个随机引物中找到了5个可用于鉴定这两个杂种纯度的引物,其中OPK14引物可同时用来鉴定两个杂种。利用该法鉴定杂种纯度不仅简单、迅速、可靠,而且因DNA的用量少,不影响被检植株的后期生长。
Genomic DNAs were extracted from young leaves of two tomato hybrids and their pa rents. PCR conditions suitable for random amplified polymorphic DNA(RAPD)analysis of tomato were studied.Of 100 arbitrary primers used in the experiment,5 primers amplified polymorphic bands between the Paris of parents. The two hybrids could be distinguished from each other using the primer OPK14.
