摘要 目的:通过人羊膜与成纤维细胞的共培养,来观察成纤维细胞在HA上的生长特性,以及胶原原纤维产生情况,为使人羊膜成为组织工程皮肤的载体打下基础。方法:将不同浓度的成纤维细胞种植于人羊膜上进行共培养,观察细胞在人羊膜细胞外基质(HA-ECM)的生长、排列以及细胞的粘附和胶原原纤维产生情况。结果:梭形的成纤维细胞在HA-ECM上生长良好,放射状或平行排列;组织切片发现HA-ECM上有较多梭形细胞附着,细胞排列紧密;电镜下见成纤维细胞胞膜基底部向HA-ECM内伸出数个突起,细胞与羊膜基质粘附相当牢固,胞外有较多胶原原纤维堆积。结论:种植于HA-ECM的成纤维细胞有一个最适宜的细胞浓度3.5×109L-1;HA-ECM具有支架和渗透作用,成纤维细胞在其上生长、粘附良好,胶原合成、分泌旺盛,HA-ECM是一种较理想的成纤维细胞载体。 AIM:To observe the growth characteristics of fibroblasts on human amnion(HAM) and the occurrence of collagen fibril by co culture of fibroblasts and HAM so as to provide a basis for the HAM as the carrier of the skin tissue engineering. METHODS:Various concentration of fibroblasts were seeded onto human amnion extra cellular matrix(HA ECM)and cultured in vitro.The performance of cells' growth, array, adhesion and collagenous fibril formation on HA ECM was observed with light microscope and transmission electron microscope. RESULTS:Fusiformed fibroblasts attached HA ECM firmly and compactly, the fusiform cells oriented radiantly or longitudinally in the trial group.A lot of fusiform fibroblasts closely packed onto the HA ECM.When viewed in the tussue slice,under EM they attached firmly and compactly.The basal membrane of fusiform fibroblasts with oval nucleus and rich in chromatin reached much protuberance into matrix of HA ECM and collagen fibril cumulating outside cells. CONCLUSION:The optimal cell concentration is 3.5×109L-1,HA ECM an is ideal carrier for fibroblasts because of its scaffold and diffusion characters.The fibroblasts attached,adhered and proliferated well on the HA ECM.
