摘要 目的 观察成纤维细胞在不同培养条件下生长 ,增殖 ,代谢及蛋白质合成等情况 ,探讨成纤维细胞的最佳培养模型 .方法 将正常皮肤中的成纤维细胞 (Ns Fb)分离出来并进行原代及继代培养 ,之后建立不同的成纤维细胞培养系统 ,分别利用绘制细胞生长曲线 ,3H-胸腺嘧啶 (3H- Td R)掺入及 3H-脯氨酸掺入等方法观察细胞的增殖 ,DNA代谢及胶原合成等情况 ,研究细胞在不同条件下的生物学特性 .结果 成纤维细胞在单层及纤维蛋白胶中培养时细胞的增殖 ,代谢及蛋白质合成等生物学功能均明显强于在两种胶原凝胶中培养时的情况 .结论 以纤维蛋白胶为支架的三维培养系统能更好地模拟生理条件下创面愈合时成纤维细胞的生物学特性 . AIM To investigate the optimum culture model of fibroblast,the difference of growth, proliferation, DNA metabolism and protein synthesis of the fibroblasts incubated in varied culture conditions. METHODS The different culture systems were established. The fibroblasts, being isolated from normal human skin samples, were cultivated and the cell growth curves were drawn. Meanwhile, 3H TdR and 3H proline incorporations were employed to measure the DNA metabolism and collagen synthesis of the cells respectively. RESULTS The biological functions of fibroblasts in bothmonolayer culture and fibrin lattice demonstrated more active and higher expressing in cell proliferation, DNA metabolism and collagen synthesis than that in the other two collagen lattices. CONCLUSION The fibroblasts in the culture system of the three dimensional fibrin lattice could be used more effectively to simulate the biological characteristics of the fibroblasts living in the wound in vivo.
