摘要
采用正交设计和响应面分析等实验方法对基因工程菌株里氏木霉 (Trichodermareesei)30 6产组织型纤溶酶原激活剂 (t PA)的液体发酵条件进行了优化。确定了适宜的培养基配方和最佳发酵工艺条件。在优化发酵条件下 ,摇瓶液体发酵液中的t PA酶活力达 3386 91IU/mL ,比初始发酵条件下酶活力提高上千倍。
The fermentation conditions of tissue-type plasminogen activator (t-PA) producing using recombinant Trichoderma reesei 306 was studied. Through orthogonal experiment and response surface methods, the optimum medium and the optimum fermentation condition were determined. The optimized medium is: bran 31 g/L, urea 14.5 g/L, tryptone 0.2 g/L, dissolubility cellulose 20 g/L, KH 2PO 4 15 g/L, MgSO 4·7H 2O 1.18 g/L, MnSO 4·H 2O 0.0013 g/L, CaCl 3 2H 2O 0.8 g/L, Tween 80 lg/L. The t-PA yield of 3386.91 IU/mL was obtained at the intial pH of 5.4, temperature of 28℃, rotation speed of 150 r/min for 36h in a 250mL flask with 50mL broth, which is over one thousand times than control.
