摘要
杏鲍菇产漆酶条件为:以20%马铃薯为基础,2%葡萄糖为碳源,0.3%酵母粉为氮源,添加5mmol/LMg2+,初始pH8.0.250mL三角瓶装液80mL,25℃,160r/min,培养9d。粗酶液性质:酶最适反应温度为55℃,最适反应pH 为4.5,1mol/L和2.5mol/LCa2+、Zn2+、K+、Fe3+对酶活性有抑制作用,5mmol/LCu2+和Mn2+对酶活性无明显影响。
The optimal conditions for laccase production by Pleurotus eryngii were studied and the results are fo lows cu ture medium made up of 20% potato, 2% glucose and 0.3% yeast power, and laccase activity can be promoted by adding 5mmol/L Mg^2+. The medium pH and temperature are 8.0 and 25℃, respectively. The fermentation lasted for 9days at 160r/min with 80mL of medium per flask. The laccase showed an optimal reaction temperature of 25℃ and pH of 4.5; Ca^2+, Zn^2+, K^+ and Fe^3+ are inhibitors for laccase and 5mmol/L Cu^2+ and Mn^2+ show no obvious inhibition.
