试纸-光电检测仪快速测定食品中亚硝酸盐的研究 被引量:19

摘要 目的利用检测试纸条和光电检测仪联用的方法 ,实现对食品中亚硝酸盐的现场快速定量检测。方法通过检测试纸与亚硝酸盐显色反应的原理 ,将显色的试纸条在光电检测仪的感应窗里测定 ,并分别通过多次实验 ,确定各种优化的实验条件。结果本方法的优化实验条件包括 :(1)显色剂的浓度为N - (1-萘基 ) -乙烯二胺 15g·L 1、对氨基苯磺酸 2 0g·L 1;(2 )样品pH为 3.5~ 5 .5 ;(3)反应时间为 6 0s ;(4 )稳定剂的浓度为酒石酸 5 0g·L 1;(5 )试纸材料为中速定量滤纸。亚硝酸盐浓度在 1.5~ 75 μg·mL 1范围内的线性关系良好 (r =0 .996 ) ;方法的检出限为 0 .4μg·mL 1。对火腿肠、香肠等样品中亚硝酸盐的含量测定 ,其相对标准偏差均 <5 % ,并与国家标准方法测定结果一致。结论试纸 -光电检测法用于亚硝酸盐的现场定量检测具有简便、快速、携带方便等特点 ,值得推广应用。 Objective To establish a rapid and accurate method for the determination of nitrite in food on the spot. Methds Based on the principle of color reaction, the content of nitrite in food was assayed by colored test strip combined with a small photoelectric meter, and the optimal experimental conditions were also determined. Results The optimal experimental conditions included (1)the concentration of color developer: N-(1-nahthyl)-ethylenediamine 15 g·L -1 , para-amino benzene sulfonic acid 20 g·L -1 , (2)pH:3.5~5.5,(3)reaction time:60 s, (4)concentration of stabilizer: tartaric acid 50 g·L -1 , (5)material of test strip: quantitative filter paper of medium velocity. There was good linear relationship between the concentrations of nitrite (in the range of 1.5~75 μg·mL -1 ) and absorbance, r=0.996; the minimal detection limit was 0.4 μg·mL -1 . The results obtained by this method for detecting nitrite in sausage or ham sausage were consistent with those by the national standard method, and the relative standard deviations were less than 5%. Conclusion Test strip combined with photoelectric meter to detect nitrite in food on the spot is a rapid, accurate method, and easy to carry, so it is worth spreading.
