摘要
观察血管内皮细胞生长因子 (VEGF)在高龄急性脑梗死 (AIS)小鼠大脑皮层内的表达及祛瘀生新中药对其的影响 ,将 2 6周龄小鼠随机分为假手术组 (A组 )、造模后 1天组 (B组 ) ,造模后 7天组(C组 )、中药 8天给药组 (D组 ) ,及中药 14天给药组 (E组 )。用电凝法在小鼠右侧脑部制备急性大脑中动脉梗死模型 ,在两个不同的时间节点取材 ,通过免疫组织化学及反转录酶聚合酶链式反应 (RT -PCR)方法来观察各组小鼠梗死区大脑皮层内VEGF蛋白及RNA的表达水平。结果 :在免疫组化方面 :E组VEGF阳性颗粒明显多于A组、B组、C组及D组 (P <0 0 5 ) ;D组VEGF阳性颗粒明显低于C组(P <0 0 5 ) ,但明显多于A组及B组 (P <0 0 5 ) ;RT -PCR方面 :E组VEGFRNA的表达明显高于D组、B组及A组 (P <0 0 1) ,D组VEGFRNA的表达明显低于C组 (P <0 0 5 ) ,但明显高于B组及A组(P <0 0 5 )。结论 :AIS小鼠梗死区内VEGF的RNA及蛋白表达会明显上调 ,并保持一段时间 ;祛瘀生新中药对AIS小鼠梗死区内VEGF的RNA及蛋白表达均具有明显促进作用 。
To observe the effect of 'Quyu Shengxin' herbs on VEGF expressions in the cerebral cortex of the old mice with acute ischemic stroke, 26-week Kuming mice were randomized into five groups: group A in which sham operation was performed, group B in which mice were dealt 1 day after modeling, group C in which mice were dealt 7 days after modeling, group D in which mice were given herbs for 8 days, and group E in which mice were given herbs for 14 days. The model of middle cerebral artery occlusion (MCAO) was established by electrocogulation on the right side of mice brain. The levels of protein granule and VEGF RNA expressions in the cerebral cortex of infarcted side were measured by immunohistochemistry and RT-PCR. Results: In immunohistochemistry, the positive granule of VEGF was more in group E than any other groups (P<0.05), less in group D than in group C(P<0.05), more in group D than in groups A and B(P<0.05); in RT-PCR, VEGF RNA expressions in were obviously higher in group E than in groups D, B and A (P<0.01), lower in group D than in group C(P<0.05), higher in group D than in groups A and B (P<0.05). It is concluded that the expressions of VEGF RNA and protein elevate in AIS mice and maintain for a period; 'Quyu Shengxin' herbs can markedly promote the elevation of VEGF RNA and protein, hence to protect and improve the infarcted cerebral cortex.
