摘要
目的 研究谷氨酸和白细胞介素 1β (interleukin 1β ,IL 1β)致痫大鼠海马组织cAMP含量的变化。 方法 40只SD雄性大鼠随机分为 4组 ,均行侧脑室注射 ,A组注射致痫剂量的L 谷氨酸钠 ;B组联合注射IL 1β和阈下剂量的L 谷氨酸钠 ;C组注射白细胞介素 1受体拮抗剂 (IL 1receptorantagonist ,IL 1ra) 3 0min后 ,再联合注射IL 1β和阈下剂量的L 谷氨酸钠 ;D组注射等容量的生理盐水。各组均在注射完毕 3 0min后断头取海马组织 ,采用放射免疫分析法(RIA)测定 4组大鼠海马的cAMP含量。结果 A、B两组大鼠均有明显的痫性发作 ,且海马cAMP的含量均明显高于D组 (P <0 0 5) ,C组大鼠海马cAMP的含量较B组下降 ,并且与D组无显著差异。结论 在谷氨酸和IL 1β的协同致痫活动中 。
Objective To investigate the changes of cyclic adenosine monophosphate (cAMP) levels in th e hippocampus of epilepsy rats induced by L-glutamate and interleukin-1β (IL-1β).Methods Forty adult SD rats were randomly divided into 4 group s (n=10 in each group). Group A was injected with L-glutamate intracere broventricularly (i.c.v.); Group B was injected with IL-1β and L-glutamat e serially in the same way; Group C was injected with IL-1 receptor antagonist (IL-1ra) 30 min followed by injection of IL-1β and L-glutamate; Group D was injected with physiological saline as control group. The animals were sacrif iced 30 min after surgery. The levels of cAMP in the hippocampus homogenate of e ach group were assayed by means of radioimmuoassay (RIA). Results The groups A and B showed epileptic activities. The cAM P levels in group A \[(593 27±172 44) pmol/g\] and group B \[(530 98±128 5 1) pmol/g\] were much higher than those in group D \[(381 19±81 28) pmol/g\] (P<0 05), while the cAMP levels in group C \[(451 95±65 98) pmol/g\] wer e decreased as compared with those in group B, and had no difference with those in group D (P>0 05). Conclusion cAMP may play a role in the onset and continuance of epileptic activities induced by IL-1β and L-glutamate.
