EFFECTS OF IL-1β OR IL-6 ON THE CELL CYCLE OF ASTROCYTES IN CEREBRAL CORTEX OF NEONATAL RATS

摘要 为研究白细胞介素1β(Interleukin1β,IL1β)和白细胞介素6(Interleukin6,IL6)对培养新生大鼠的大脑皮质星形胶质细胞增殖的影响,本研究采用体外细胞培养方法获得新生大鼠大脑皮质星形胶质细胞,然后在培养液中分别加入IL1β和IL6,在作用6、12及24h后,通过流式细胞仪检测IL1β、IL6对星形胶质细胞细胞周期的影响。结果显示IL1β、IL6作用于星形胶质细胞后,可引起细胞周期的显著变化,表现为G1期细胞数减少,S期、G2/M期细胞数增多,以作用12h时变化最明显。提示IL1β、IL6可促进体外培养的星形胶质细胞增殖。 To study the effects of IL-1β or IL-6 on the proliferation and cell cycle of cultured cerebral cortex astrocytes. In the present study, cerebral cortex astrocytes of neonatal rats were generated through cell culture. IL-1β and IL-6 were administrated to cultured astrocytes. The effects of IL-1β and IL-6 on the cell cycle of astrocytes were detected by means of flow cytometer at 6 h, 12 h or 24 h following drug application. The results showed that significant changes of cell cycle occurred following administering IL-1β or IL-6. The percentage of cells in G1 cycle phase was reduced and that of S and G2/M cycle phase increased, with the effects on cell cycle being the most significant at 12 h post drug application. These findings suggest that IL-1β and IL-6 may promote the proliferation of cultured cerebral cortex astrocytes.
