摘要
对影响马铃薯花粉超低温保存效果的因素进行了研究。结果表明:冷冻前干燥处理18 h的花粉,保存后其生活力优于干燥处理12 h和24 h的花粉;用0℃(12 h)→-10%(12 h)→-20℃(12 h)的逐步降温预冻处理,可大幅度提高在液氮中冰冻保存花粉的活力;保存后的花粉用-20℃(12 h)→4℃(12 h)→25%(12 h)逐步解冻效果最好,35~40℃直接解冻效果优于(25±2)℃直接解冻;超低温保存对花粉萌发表现出某些促进作用。
The factors of affecting the viability of potato ( Solarium tuberosum) pollen stored in liqiud nitrogen (LN) were investigated in order to improve cryopreservation effect. The results indicated that desiccation . duration of 18 h and pre-freezing treatment at varied temperature of 0℃ (12 h) →- 10℃(12 h)→- 20℃ (12 h) can greatly increase the viability of pollen stored in LN before thrown into LN. The method of step by step defrozen of - 20℃ (12 h) →4℃ (12 h) →25℃ (12 h) is better than that of defrozing pollen directly in 35 - 40℃or in (25±2)℃; The cryopreservation enhances pollen germination of some varieties.
