摘要
对新疆莎车县扁桃品种晚丰18号花粉进行了干燥法和玻璃化法2种超低温保存方法研究,结果表明:干燥方式对晚丰18号扁桃花粉超低温保存后的萌发率有显著影响,其中以4℃硅胶干燥最好,干燥脱水时间应控制在8 h左右,花粉含水量为52.74%,经超低温保存后花粉萌发率可达65.45%,且花粉率与保存时间无关;以4℃2 h、室温(20±1)℃30 min和35℃70 s 3种方式解冻,解冻后花粉萌发率差异极显著,其中以35℃70 s的萌发效果最好,解冻后花粉萌发率达63.09%;干燥法保存效果较玻璃化法保存好。
The pollen of almond cuhivars wanfeng-18 was tion. The results showed that:effect of germination rate of preserved at low temperature by drying and vitrificadrying methods on the wanfeng-18 of almond pollen cryopreservation was significant, the silica gel drying at 4 ℃ was best, the drying time should be controlled around 8 h, pollen water content of 52.74% ,the eryopreservation of pollen germination rate reached 65.45%, and pollen rate and save time independent;Thaw in 3 ways at 4 ℃ 2 h,the room temperature (20 ± 1 )℃ 30 min and 35 ℃ 70 s, after thawing pollen germination rate significantly, the 35 ℃ 70 s had the best effect of germination, after thawing pollen germination rate reached 63. 09% ; Drying preservation was better than vitrification preservation.
