摘要
采用羟丙基 β 环糊精 (HP β CD)作为手性选择试剂对外消旋的苯磺酸氨氯地平进行了拆分 ,研究了环糊精种类 ,浓度 ,缓冲液的pH值以及添加剂对分离的影响 ,结果表明以羟丙基 β 环糊精为手性选择剂 ,短链的阳离子表面活性剂四乙基氯化铵为电渗流改性剂可以使苯磺酸氨氯地平实现基线分离 。
Amlodipine was separated by capillary electrophoresis using hydroxypropyl-β-cyclodextrin as chiral selector. The effects of type and the concentration of cyclodextrins, pH of the buffer used on the resolution were investigated. The effect of cationic surfactant was also examined. Good resolution of amlodipine was achieved using phosphate at pH 3.0 containing 5mmol/L hydroxypropyl-β-cyclodextrin, 50mmol/L Tetraethylammonium-chlorid and 2mol/L urea. Amlodipine can be enatiomerically resolved in less than 22 min with no capillary treatment. The mechanism of chiral separation with β-cyclodextrin and hydroxypropyl-β-cyclodextrin.
