摘要
用MTT显色方法对体外传代K562,SW1116,Bcap-37和HR8348细胞分别进行了抗癌药物敏感性、抗癌药物调变,激素和激光光动力学作用的研究,均得到较稳定的结果。各项研究带有共性的结果是K562等细胞在一定范围内与理化作用的强度及作用时间长度呈正相关。因而,在实际应用中,可根据不同的实验对象和目的,对MTT法的实验条件作各种特定性设计。本文提出成功的MTT试验对照孔OD值宜超过0.5。并就理论最高血药浓度C_(max)、1/10 C_(max)及由Cmax降至30%抑瘤时药物浓度的倍减次数的价值进行了讨论。
This paper reports the application of MTT assay to study on the inhibitory effect of chemotherapeutic drugs, chemotherapy modulator, sex hormone and laser photodynamic treatment for various human cancer cell lines, with satisfactory results. Within a certain range exist positive correlationships between the strengh of the physical or chemical factors and the length of the exposure time. Exception was also found in little time-dependent effect during the 2-5 day continuous drug exposure for Hom, MTX, Fudr and DNR. As a general 2-day exposure MTT assay, it is fit for special designs to meet the experimental purposes. In this paper the authors discussed the theoretical maxmal plasma drug concentration(Cmax) and application of the 1/10 Cmax to drug sensivity tests. The authors also had a discussion on the value of deductable times of drug concentration from the Cmax to the concentration at which a 30% inhibitory effect was reached.
