摘要
目的:观察地塞米松对哮喘大鼠模型气道壁胶原、转化生长因子β_1(TGF-β_1)含量的影响。方法:30只SD大鼠随机分成正常对照组、哮喘模型组和激素干预组,以卵蛋白致敏并长期吸入激发制备大鼠慢性哮喘模型,干预组于每次吸入前腹腔注射地塞米松,模型组以生理盐水代替。肺组织石蜡切片行胶原和TGF-β_1免疫组化染色后计算机图像分析测定其含量。结果:模型组气道壁Ⅲ型胶原及TGF-β_1含量均高于对照组(q值分别为13.57、12.76,P值均<0.001);激素于预组Ⅲ型胶原及TGF-β_1含量低于模型组(q值分别为12.88、11.23,P值均<0.001),较之对照组差异均无显著性;各组间Ⅰ型胶原的含量差异不大(P值均>0.05)。TGF-β_1的表达与Ⅲ型胶原的含量呈显著正相关(r=0.606,P<0.01)。结论:地塞米松可以减少气道壁Ⅲ型胶原的沉积和TGF-β_1的表达,对Ⅲ型胶原沉积的抑制可能部分通过抑制TGF-β_1表达而实现的。
Objective: To observe the effect of desamethasone (DXM) on the collagen deposition and expression of transforming growth factor-pi (TGF-pi) in asthmatic rats. Methods: Thirty SD rats were randomly divided into 3 groups: the normal group, the model group, the treatment group, with 10 rats in each group. The asthmatic model was prepared by repeated inhalation of ovalbulium. The alteration of collagen and TGF-b2 content in the airway wall were measured by the computerized image analysis system. Results: The contents of collagen type HI and TCP-Pi in airway wall were significantly higher in the model group than those in the normal group( q = 13. 57,12. 76 respectively, P < 0. 001). In the treatment group, the contents of collagen type III and TCP-Pi in airway wall were lower than those in the model group( q = 12. 88, 11. 23 respectively, P < 0. 001), and there wes no difference between treatment and normal group(P> 0. 05). The contents of collagen type I in airway wall has no diffrence among the 3 groups ( P > 0. 05) . There was a close correlation between TGF-b1 and collagen type Iff ( r. = 0. 606, P < 0. 01) . Conclusion: DXM may reduce the precipitation of collagen and inhabit the the expression of TGF-b1 in airway wall.
