阿托伐他汀对大鼠心肌梗死后心肌间质转化生长因子-β1表达的干预研究 被引量:2

摘要 目的探讨大鼠心肌梗死后心肌间质转化生长因子-β1(TGF-β1)的表达及阿托伐他汀对其的影响。方法结扎SD大鼠冠状动脉前降支复制急性心肌梗死模型,随机分成心肌梗死对照组(n=26)和阿托伐他汀治疗组(n=27)。两组又随机分为1周、2周和4周亚组。另设假手术组(n=8)。用逆转录-聚合酶链反应法(RT-PCR)及免疫组化方法检测TGF-β1 mRNA和蛋白表达情况。结果与假手术组相比,心肌梗死对照组TGF-β1的mRNA表达在各亚组明显升高(均P〈0.05),1周和2周亚组蛋白表达也均增高(均P〈0.05)。而阿托伐他汀治疗组TGF-β1蛋白表达在1周和2周亚组,mRNA的表达在1周、2周、4周亚组,均较心肌梗死对照组显著降低(均P〈0.05)。结论阿托伐他汀可抑制心肌梗死后心肌间质TGF-β1的表达,可能会间接改善心肌梗死后心室重塑的发生发展过程。 Objective To investigate the effect of atorvastatin on expression of transforming growth factor - betal ( TGF - β1 ) of myocardial extracellular matrix after infarction in the rats. Methods One hundred and ten adult male Sprague - Dawley rats underwent the left descending coronary artery ligation , then being divided randomly into AMI control group (n = 26)and AMI atorvastatin treatment group (n = 27). The rats in those two groups were redivided into 3 sub - groups according to the course of treatment : 1 - week, 2 - week, 4 - week sub - groups. Eight rats underwent sham operation, being killed after 4 weeks. The mRNA and protein expression of TGF - β1 was examined by reverse transcription - polymerase chain reaction ( RT - PCR) and immunohistochemistry. Results Compared to sham operated rats, the mRNA expression of TGF - β1 showed significant increase in all sub- groups of AMI controls( all P 〈0.05), and the protein expression of TGF - β1 were enhanced in the 1 - week and 2 - week sub - groups ( all P 〈 0.05 ) . In comparison with the AMI control group, the protein expression of TGF - β1 were significantly lower in the 1 - week and 2 - week sub- groups of atorvastatin group(all P 〈0.05) and so were the mRNA expression of TGF- β1 in the 1 - week,2 - week,4 - week sub - groups ( all P 〈 0.05 ). Conclusion The expression of TGF - β1 in the myocardial extracellular matrix increased after infarction. These changes suggest that TGF - β1 plays an impor- tant role in postinfarction extracellular matix remodeling. Atorvastatin probably can prevent ventricular remodeling by suppressing the enhanced expression of TGF - β1.
