摘要
【目的】通过对卵裂期胚胎培养液可溶性人类白细胞抗原G(sHLA-G)的定性和定量分析,了解卵裂期胚胎sHLA-G抗原表达及其与胚胎发育和种植的关系。【方法】应用免疫发光WesternBlot了解卵裂期胚胎sHLA-G和HLA-I类抗原的表达情况;应用双抗体夹心法ELISA了解卵裂期胚胎培养液中sHLA-I类抗原的表达量与胚胎发育和种植的关系。【结果】①胚胎培养液中以sHLA-G抗原为主,其它sHLA-I类抗原量极少或不存在,因此用培养液中sHLA-I类抗原的量来估计培养液中sHLA-G的含量是可行的。②卵裂期胚胎培养液中1级胚胎sHLA-I类抗原的绝对吸光度(A)值为0.077±0.006;2级胚胎为0.064±0.002;3级胚胎为0.043±0.002;3组比较差异有显著性(P=0.014)。胚胎种植率与胚胎sHLA-I类抗原绝对A值的相关系数为0.426,P=0.004。它们的表达量与胚胎发育和种植的有关。【结论】卵裂期胚胎培养液中sHLA-G的表达量与胚胎发育和种植的有关。
Objective] To study soluble human leukocyte antigen G(sHLA G) expression of cleavage stage embryo and the relationship between SHLA G embryo development or implantation by detecting the quality and quantity of sHLA G in their culture medium. The expression of sHLA G and other sHLA I antigens were detected by Immunofluorescence Western Blot. sHLA I levels were measured in each embryo's culture medium by ELISA and the relationships between sHLA I antigens and embryo development and implantation of cleavage stage embryo were analyzed. ① Western blot analysis showed dominant expression of soluble HLA G antigen. Other soluble HLA I antigens were not detected by the same method. Therefore the level of HLA I in culture medium was close to the level of HLA G. ② The value of soluble HLA I or sHLA G secreted from the first class cleavage stage embryos was 0.077± 0.006, the second class embryo was 0.064± 0.002, the third class embryo was 0.043± 0.002, and there were significant differences in the three groups (P=0.014). Correlation coefficient of the A value of sHLA I and embryo implantation rate was 0.426, P=0.004. So the soluble HLA I or sHLA G secreted from the cleavage stage embryos was related to embryo implantation rate and embryo quality. [ Conclusion] sHLA G expression is closely related to embryo development and implantation of cleavage stage embryo.
