摘要
对丽格海棠叶片、叶柄离体培养及快速繁殖方法进行研究。添加0.5mg/L2,4-D和0.5mg/LKTMS培养基上培养的外植体,首先形成愈伤组织,将其转移到不含激素的MS基本培养基上后,先后形成不定根和不定芽,其不定芽分化频率为85%以上。培养在添加0.2mg/LNAA和2.0mg/LBAPMS培养基上的外植体,无愈伤组织形成,培养2周后开始直接形成丛生芽。当转移到MS基本培养基上后,长成小苗,其再生频率为90%以上。再生植株移栽于蛭石+草炭土基质中,成活率90%以上,并至开花。
Efficient plant regeneration in leaf and petiole explant cultures of Begonia×Elatior was studied. Explants produced calli on the MS induction medium supplemented with 0.5 mg/L 2,4-D and 0.5 mg/L KT. When transferred onto the hormone-free MS medium, over 85% of the calli regenerated shoots. The explants formed shoots after 2 weeks cultured on the MS induction medium supplemented with 0.2 mg/L NAA and 2.0 mg/L BAP. The shoot regeneration frequencies reached up 90%. When grew in natural condition, the regenerated plants flowered.
