摘要
把经过纯化的香菇单核营养缺陷型(?)原生质体,与碘代乙酰胺灭活的野牛菇 L_(81-39)_4,原生质体用PEG 进行融合,在双层再生培养基上培养,挑得融合子5株:F_4,F_6,F_(11),F_(17),F_(18)。用荧光染色法,对亲本及融合子菌丝的单核和双核进行详细观察和鉴定.融合子 F_4,F_6,F_(18) 出菇表明,菇形偏近 L_(81-39),但菌柄较短,菌盖较大且略厚;F_(17)的菇色,肉质程度偏向于 L_(52);F_(11)出菇多,但个体小,色淡,菌盖薄。酯酶同工酶测定结果表明:F_(11),F_(17)都有双亲所具有的9条谱带中的7条,R_f值为:0.683,0.792,0.833,0.850,0.875,0.892,0.917.F_4缺少 F_(11)和 F_(17)所具有的 R_r=0.683的谱,F_6缺少 R_f=0.683和R_f=0.850的谱带.融合子都没有表现出 L_(81-39)的 R_f=0.733,L_(52)的 R_f=0.667这二条谱带。此外,融合了都有一条双亲没有的 R_f=0.808的谱带。
Five fusants(F_4,F_6,F_(11),F_(17),F_(18))were obtained from the fusion of protoplasts of parent strainsby the polyethylene glycol-Ca^(2+),two parent strains used were adenine auxotrophic mutant L^(52)^(-ade)andL_(81-39),which lost regeneration activity and were treated with iodoacetamide.The homokaryons orheterokaryons of parent.strains and fusants were observed and identified by fluorescent nuclear straining.The mycelium growth rate and fruitbody formation were compared between the fusants and parent strains.Fruit bodies of the fusants F_4,F_(18)have gcneral appearance of those from parent strain L_(81-39),withshorter stipes and thicker caps.While F_(17)has abnormal shape,but the colour and thickness of the caps weresimilar to parent strain L^(52)^(-ade).F_(11)has smaller and thinner caps.The isoenzyme of esterase of the fusants wasidentified by diseontinuos polyacrylamide glycerol electrophosis.
