摘要
应用正交设计法研究了不同因素对茯苓原生质体制备的影响.实验结果表明,渗稳剂的种类和酶解时间对茯苓原生质体产量的影响最显著;培养5 d的菌丝体在30 mg/L溶壁酶+20 mg/L蜗牛酶的酶液中,以0.6 mol/L的蔗糖作为渗稳剂,30℃酶解1 h,茯苓原生质体的产量最高.
The effects of different factors on Poria cocos protoplast preparation were studied with orthogonal design method. The experimental results showed that both the category of osmotic pressure stabilizer and the time of enzyme treatment acted on Poria cocos protoplast yields were the most distinct, and in the conditions of the mycelium enzyme cultivated for 5 days in 30 mg/L Lywallzyme and 20 mg/L Snailase at 30 ℃ for 1 hour with 0. 6 mol/L sucrose as osmotic pressure stabilizer, the Poria cocos protoplast yield was the highest.
