摘要
目的 :应用高效液相色谱法测定血液中甲氨蝶呤药物浓度 ,以指导临床用药。方法:采用NucleosilC18钢柱 ,甲醇/磷酸盐缓冲液 (16/84)为流动相 ,紫外检测波长306nm ,微量血中 ,高氯酸沉淀蛋白 ,少量氢氧化钠调节酸度后 ,上清液进样分析。结果 :MTX及代谢产物色谱分离理想 ,在0 1~20.0mg/L浓度范围内线性关系良好 ,相关系数为0 9999,在高、中、低4种浓度下 ,平均相对回收率为102 1%、101 2 %、101 4 %、99 48 % ,平均绝对回收率为94 68 %、94 74 %、91 89%、90 48% ,日内精密度为3 85%、2 59%、3 38%、3 20 % ,日间精密度为4 95 %、6 03 %、5 60 %、3 98% ,最低检出限为0 05mg/L血清。结论 :该方法适用于儿科临床血药浓度监测 。
Objective:To establish high performance liquid chromatographic method for determining the blood methotrexate(MTX)concentrations.Methods:With Nucleosil C 18 column,the mobile phase was consisted of16%methanol and84%phosphate buffer.Protein was precipitated from serum sample with perchloric acid.Supernatant with a few sodium hydroxide was analyzed at uv306nm by the mobile phase.Results:The satisfied MTX chromatographic peak was showed.The good linearity was obtained at the range of0.1~20.0mg/L(r=0.9999).At four concentrations,the average recoveries were102.1%,101.2%,101.4%,and99.48%,reˉspectively.RSD of daytime were4.95%,6.03%,5.60%and3.98%,respectively.The minimal detectable conˉcentration was0.05mg/L.Conclusion:This method is practical for monitoring the blood MTX concentration and can provide the basis for clinical effective and safe treatment.
