摘要 目的 探讨高脂肪饮食对胰岛素水平及脂肪组织脂联素mRNA表达的影响。方法 成年小鼠随机分 2组 ,一组 (n =2 5 )予普通饮食 ,另一组 (n =2 5 )予高脂肪饮食 ,12周后取血测血浆胰岛素和血糖浓度 ,取皮下脂肪组织 ,提取脂肪组织总RNA ,RTPCR检测脂联素mRNA的表达。结果 高脂饲养组鼠体重量、血糖未增加 ,脂肪量和血胰岛素水平均较普通饲养组升高 (P <0 0 1)。脂联素mRNA表达量在高脂饲养组减少 (P <0 0 5 ) ,与胰岛素水平呈负相关 (r =- 0 771,P <0 0 1)。结论 高脂饮食升高胰岛素水平 ,并减少脂联素mRNA的表达。 Objective To explore the effect of high fat diet on insulin concentration and adiponectin mRNA expression in adipose tissue in house mice. Methods Mice were randomly divided into two groups, and were fed respectively by normal and high fat diet. Plasma insulin and glucose were measured after 12 weeks. Total RNA was extracted from adipose tissue. The adiponectin mRNA expression was tested by RT-PCR. Results No significant difference in body weight and plasma glucose was not significantly observed between the two groups. Fat mass and plasma insulin concentration were higher in high fat diet group than in normal diet group(P<0.01). But the expression level of adiponectin mRNA was lower in high fat diet group than in normal diet group(P<0.05).There was a negative correlation between insulin concentration and expression level of adiponectin mRNA in adipose tissue(r= -0.771, P<0.01). Conclusion High fat diet increases insulin concentration and decreases adiponectin mRNA expression in mice.
