摘要 应用RAPD标记鉴定番茄杂交种子纯度,从160条随机引物上筛选出BS273引物,能在杂交一代父母本中各自产生特异的RAPD标记,并且父母本的特异标记能在杂交一代中同时表现,从而利用一个引物便可将杂交一代及其父本、母本三者清楚地区分开,同时也能鉴别其它混杂种子。结果表明:利用RAPD标记技术鉴定番茄杂交种子纯度有着快速和准确的优势,对于杂交种子生产具有广阔的应用前景。 RAPD markers were applied to determine the purity of tomato F1 hybrid seeds with total genomic DNA in this study. The primer BS273 was screened out of 160 arbitrary 10-mer oligonucleotide primers, which generated distinctive bands between male, female parents and hybrids. The specific bands of parents could be expressed in F1 hybrids, male, female parents and hybrids could be distinguished clearly by one primer and other mixed seeds could be detected too. The results showed that such a technique was rapid, accurate and had a significant application future in the production of tomato hybride seeds.
