摘要
应用RAPD标记鉴定番茄杂交种子纯度,从160条随机引物上筛选出BS273引物,能在杂交一代父母本中各自产生特异的RAPD标记,并且父母本的特异标记能在杂交一代中同时表现,从而利用一个引物便可将杂交一代及其父本、母本三者清楚地区分开,同时也能鉴别其它混杂种子。结果表明:利用RAPD标记技术鉴定番茄杂交种子纯度有着快速和准确的优势,对于杂交种子生产具有广阔的应用前景。
RAPD markers were applied to determine the purity of tomato F1 hybrid seeds with total genomic DNA in this study. The primer BS273 was screened out of 160 arbitrary 10-mer oligonucleotide primers, which generated distinctive bands between male, female parents and hybrids. The specific bands of parents could be expressed in F1 hybrids, male, female parents and hybrids could be distinguished clearly by one primer and other mixed seeds could be detected too. The results showed that such a technique was rapid, accurate and had a significant application future in the production of tomato hybride seeds.
