摘要
用正交试验法确定胰蛋白酶酶解文蛤的最佳工艺条件 ,用薄层层析和氨基酸自动测定仪鉴定酶解液中氨基酸的种类和含量 ,进一步判定酶解效果。结果表明 :在 5 0℃ ,加酶量 2 %,pH8.0 ,酶解 8h的酶解效果最好。层析发现 ,酶解温度为5 0℃的提取液含氨基酸较全面 ,氨基酸自动测定仪测定显示 ,胰蛋白酶酶解法的文蛤提取液中氨基酸的含量大于热水提取法的含量。
The best technological condition of digesting Meretrix meretrix was determined by method of orthogonal experiment. The result showed that the best effect of enzyme was in the condition of 50℃, 2% of trypsin, pH8.0~8.5, and 8 hours for digestion. We discovered that the content of amino acid was more overall at 50℃ by TLC. The amino acid analyzer showed that the content of amino acid in extracting solution was more than that in hot water.
