摘要 目的观察回转器模拟失重对ROS1 7/2 .8(大鼠骨肉瘤 )细胞分化相关基因表达的影响。方法对培养的ROS1 7/2 .8细胞采用回转器模拟失重 2 4、48及 72h ,并设 1G对照组。提取细胞总RNA ,进行反转录PCR ,检测I型胶原α1链和碱性磷酸酶mRNA的表达量 ,计算与内参照物 β actin的比值。结果模拟失重 72h后 ,失重组细胞内COL Iα1及alklinephosphatase(ALP)的基因表达量明显低于对照组 (P <0 .0 1 )。结论回转器模拟失重 72h使大鼠骨肉瘤细胞的分化功能降低。 Objective To investigate the effects of rotating clinostat simulated weightlessness on the differentiation related gene expression of ROS17/2.8(rat osteosarcoma) cells. Method Cultured ROS17/2.8 cells were exposed to rotating clinostat simulated weightlessness for 24, 48 and 72 h with control group in 1 G condition. Total RNA in cells was isolated and reverse transcription PCR analysis was made. Gene expression of type I collagen α1 chain and alkaline phosphatase were examined by their ratios to β actin. Result The expression of type I collagen α1 chain and alkaline phosphatase(ALP) gene was significantly lower after 72 h exposure to clinostat simulated weightlessness than that of the control group (P<0.01).Conclusion Differentiation of rat osteosarcoma cells was reduced after exposure to rotating clinostat simulated weightlessness for 72 h.
