摘要
目的 探讨空间骨丢失的机理 ,研究模拟失重时人成骨样细胞细胞周期的变化。 方法 利用回转器模拟失重 ,观察测定了成骨样细胞增殖、周期的变化及其恢复作用和中药三花接骨散对细胞周期的影响。 结果 模拟失重作用后 ,成骨细胞增殖受到明显抑制 ,G1 期细胞显著增多 ,S与 G2 +M期细胞显著减少。模拟失重 12 h后再经 12 h以上正常生长 ,细胞周期可恢复至对照水平。三花接骨散能促进 G1 期细胞减少 ,S与 G2 +M期细胞增多 ,具有对抗失重效应 ,促进成骨细胞增殖的功效。 结论 模拟失重抑制成骨样细胞增殖 ,抑制细胞 G1 期向 S期的转化 ;模拟失重作用后细胞周期有一定的恢复作用 ;药物可以刺激细胞周期对抗模拟失重的效应。
Objective To investigate the effects of simulated weightlessness on osteoblast like cell cycle and the way to protect human body against bone loss during space flight. Methods The rotating clinostat was used to simulate weightlessness .The proliferation and cell cycle of cultured osteoblast like cells in the condition of simulated weightlessness were observed and compared with those of controls.The after effect of simulated weightlessness for various hours and the effect of adding Sanhua(a kind of traditional Chinese medicine ) into the culture medium on cell cycle were also observed. Results 1. The proliferation of osteoblast like cells was inhibited significantly after 24 h of simulated weightlessness. 2. Under simulated weightlessness,the cells in G 1 phase were significantly higher than those of controls,while the cells in S and G 2+M phase were significantly less than controls.3. After rotating for 12 h, the cell cycle could restore to control's level following culture in stationary condition for 12 h or more.4. Sanhua was helpful for decreasing the cells in G 1 phase and increasing the cells in S and G 2+M phase under simulated weightlessness. Conclusions Simulated weightlessness can inhibit the conversion of cell from G 1 phase to S phase, so that the proliferation of osteoblasts was retarded. It is suggested that Sanhua may have protective effect against bone loss during space flight.
