中药复方含药血清对回转模拟失重成骨细胞影响的观察

Effects of Serum containing Traditional Chinese Compound Medicine on Osteoblast under Clinorotation-induced Weightlessness

摘要 目的:观察中药复方含药血清对模拟失重条件下体外培养成骨细胞的影响,探讨该方对抗骨丢失的部分作用机理。方法:分离乳鼠颅骨成骨细胞,体外培养后,随机分为5%,10%,20%浓度的空白血清对照组和5%,10%,20%空白血清回转组和5%,10%,20%含药血清回转组,连续回转48h模拟失重,观察该方对大鼠成骨细胞增殖、I型胶原合成、碱性磷酸酶(ALP)和骨钙素(BGP)生成的影响。结果:5%和10%空白血清回转组成骨细胞增殖比正常重力组明显降低(P<0.05);与10%空白血清回转模拟失重组相比,10%含药血清回转组可显著促进细胞增殖(P<0.05)。正常重力组与回转模拟失重组Ⅰ型胶原含量比较差异无统计学意义(P>0.05);与5%和10%空白血清回转模拟失重组相比,5%和10%含药血清回转组均能明显促进成骨细胞Ⅰ型胶原合成(P<0.05)。与5%正常重力组相比,5%空白血清回转组成骨细胞ALP活性明显降低(P<0.05);与5%和20%空白血清回转模拟失重组相比,5%和20%含药血清回转组能明显提高成骨细胞ALP活性(P<0.05)。与10%、20%空白血清组相比,10%、20%空白血清回转组成骨细胞BGP含量明显降低(P<0.05);而中药复方含药血清对其模拟失重成骨细胞BGP活性没有显著影响(P>0.05)。结论:该方能改善模拟失重条件下成骨细胞增殖,分化成熟,增加胶原蛋白合成,从而实现对抗失重骨丢失的效果。 Objective:To study the effects of serum containing Chinese compound medicine on osteoblast under clinorotation --induced weightlessness (CIW), so as to explore its mechanism of anti--bone loss. Mcthods:Osteoblasts were separated from cranial bones of neonate rats and cultured in vitro. The groupings were as follows: serum treated groups with the con- centrations of 5%, 10% and 20%, serum and CIW treated groups with 3 concentrations as above, and serum containing Chinese medicine compound plus CIW (48h) treated groups with the above 3 concentrations. The proliferation, synthesis of collagen type Ⅰ , alkaliphosphatase (ALP) and bone gla protein (BGP) of osteoblasts were observed. Results:Compared with cells in normal weight environment, osteoblast proliferation was obviously decreased in 5 % and 10 % serum groups (P〈%0.05). Osteoblast proliferation was remarkably promoted in 10% serum containing medicine plus CIW group compared with 10% serum plus CIW group (P〈0.05). There was no significant difference of type Ⅰ collagen between CIW group and normal group. Compared with 5% and 10 % serum and CIW groups, 5 % and 10% serum containing Chinese medicine compound plus CIW groups markedly improve the synthesis of type Ⅰ collagen in osteoblasts (P〈0.05). Compared with 5% normal weight group, ALP activity in 5% serum and CIW group was statistically decreased (P〈0.05). Compared with 5% and 20% serum and CIW groups, 5% and 20% serum containing Chinese medicine compound plus CIW groups remarkably increased ALP activity in osteoblasts (P〈0.05). Compared with 10% and 20% serum groups, the BGP content of osteoblasts in 10% and 20% serum and CIW groups obviously decreased (P〈0.05). However, serum containing Chinese medicine had no effect on BGP content in CIW osteoblasts (P〉0.05). Conclusion: The formula can improve the proliferation, differentiation and collagen synthesis of osteoblasts in CIW environment, so as to inhabit bone loss in weight- lessness.
