摘要
目的 :探讨HSP70 在链脲佐菌素 糖尿病性白内障发生、发展中的作用。方法 :将 6 0只SD大鼠随机分为两组 ,正常对照组与白内障组。用链脲佐菌素 (STZ)诱发糖性白内障 ,每周观察晶状体的变化 ,在实验开始后 2w末、4w末、8w末 ,分别摘取眼球 ,检测热休克蛋白 70 (HSP70 )在晶状体上皮细胞 (LECs)中的表达情况。结果 :对照组晶状体一直保持透明 ,白内障组晶状体在 2w末出现空泡 ,8w末全部混浊。HSP70 在对照组中未见表达 ,在白内障组中表达明显 ,并随着白内障的发展而增加。结论 :HSP70 可能通过调节LECs的生长在糖性白内障的发生、发展中起重要作用。
Objective:To evaluate the role of heat shock protein(HSP 70 ) in the pathogenesis of streptozotocin(STZ) diabetic cataract.Methods:60 SD rats were randomly divided into two groups:normal control and cataract. Diabetic cataract was induced by STZ intraperitoneal injection(70 mg/kg in 0.1 mol/L sodium citrate buffer). Lenses were examined once a week with a slit lamp. Eyeballs were isolated at the end of 2 weeks,4 weeks,and 8 weeks after STZ injection. Immunohistochemical staining was used to detect the expression of HSP 70 in normal and diabetic LECs.Results:The lenses of the control group maintained complete transparency throughout the experimental period;the lenses of the cataract group began to exhibit a vacuolar stage by week 2 and near opacity by week 8. The expression of HSP 70 was significantly different between normal and diabetic LECs,and was correlated with the development of cataract.Conclusion:HSP 70 may play a critical role in the pathogenesis of diabetic cataract by modulating the growth of LECs.
