摘要
目的探讨热休克蛋白(HSPs)70在年龄相关性白内障晶状体上皮细胞(LECs)中的表达及意义。方法采用Western blot蛋白印迹法,检测68例(其中皮质型23例、核型29例、后囊下型16例)年龄相关性白内障前囊膜上皮细胞中HSP70的表达,以8例正常成人晶状体前囊膜上皮细胞中HSP70含量为对照。结果HSP70在年龄相关性白内障(1.454±0.241)和正常人(0.713±0.009)LECs中的表达差异有统计学意义,年龄相关性白内障皮质型、核型、后囊下型HSP70表达也不相同。结论HSP70在年龄相关性白内障的发生、发展中可能起一定的细胞保护作用,HSP70在年龄相关性白内障皮质型、核型、后囊下型的作用机制可能不完全相同。
Objective This study was to investigate the expression of heat shock protein 70(HSP70) in lens epithelial cells of human age-related cataract and clarify the effect of HSPTO on pathogenesis of age-related cataract. Methods Sixtyeight specimens of lens capsule from cataract patients with cortical cataract(23 cases) ,nuclear cataract(29 cases) and posterior subcapsular cataract( 16 cases) were collected during the surgery, and 8 specimens of normal lens capsule from healthy corpse eyes were used as control. The expression of HSP70 in lens capsule was assayed using Western bolt. Results The expression of HSP70 was significantly higher in lens epithelial cells with cataract( 1. 454 ±0. 241 ) than in transparent lens (0. 713 ±0. 009 ) (P 〈0. 01 ). Among the different types of cataract, the expression of HSP70 was significantly higher in nuclear cataract than in cortical and posterior subcapsular cataract ( HSP70/actin: 1. 768 ±0. 053 versus 1. 758±0. 081 and 0. 836 ±0. 042 ) ( P 〈 0. 01). Conclusion Overexpression of HSP70 is associated with the development of age-related cataract, and age-related cortical cataract,nuclear cataract and posterior subcapsular cataract may possess a different pathogenesis.
