摘要
目的:探讨糖尿病发病初期大鼠发生性与生殖功能障碍的细胞学机制。方法:通过给成龄雄性大鼠腹腔注射链脲菌素制成糖尿病动物模型,分别观察1d、3d、5d、7d、14d。观察期满后断头处死,取睾丸。应用常规组织学方法观察睾丸组织形态的改变;应用流式细胞分析术检测睾丸间质细胞、支持细胞及各级生精细胞的细胞周期的改变。结果:睾丸间质细胞及生精细胞的形态、数目从致病d7开始有明显变化,支持细胞的形态无明显变化;睾丸中各类细胞在高糖环境下24h即发生增殖减慢、细胞滞留在G1期、DNA合成减少等改变。结论:糖尿病大鼠生殖功能障碍与睾丸间质细胞、生精细胞的形态、数目变化及睾丸各类细胞的增殖改变有关。
Objective: To study cytological mechanism of sexual and reproductive malfunction of rats at the initial stage of diabetes. Methods: Adult male Wistar rats were introperitonally injected with steptozocin to set up animal model of diabetes. The rats were observed at 1 d, 3d, 5d, 7d and 14 d respectively. Then the animals were killed and the testes were removed. Testicular morphological changes were observed by using regular histology method. Cell cycle changes of Leydig cells, Sertoli cells and spermatogenic cells were examined by using flow cytometry (FCM). Results: Significant changes in morphology and number of Ley-dig cells and spermatogenic cells occured from the 7th day of diabetes. Morphology of Sertoli cells did not changed significantly. On the 24th of high-glucose, decrease of proliferation speed of testes cells occured, stagnated in stage G1 and DNA synthesis were diminished. Conclusion: Reproductive malfunction of rats with diabetes is correlative with changes of morphology and number of testes Leydig cells and spermatogenic cells, and correlative with changes of testes cell proliferation.
