摘要
目的研究山茱萸总皂苷对链脲佐菌素(STZ)致糖尿病大鼠睾丸病变的干预作用。方法雄性SD大鼠一次性腹腔注射STZ 60 mg/kg造成糖尿病模型。28 d后将血糖高于16.7 mmol/L的大鼠随机分为:模型组、山茱萸总皂苷组(口服,80 mg/kg)、甲基睾酮组(口服,3 mg/kg),另加正常大鼠作对照组,每组12只。药物以0.5%CMC-Na溶液混悬后灌胃。正常和模型组以CMC-Na溶液灌胃。连续28 d。处死动物,取睾丸、附睾、精囊腺精密称重,计算脏器指数。取血清检测血糖、睾酮,取睾丸检测标志性酶乳酸脱氢酶(LDH)、酸性磷酸酶(ACP)、谷胺酰转肽酶(γ-GT),并作组织切片,H.E染色,置光镜下观察病理变化。结果糖尿病大鼠相对正常组血糖升高、睾酮水平下降,睾丸中LDH、ACP、γ-GT活性减弱。睾丸曲细精管萎缩,各级生精细胞减少,排列疏松。山茱萸总皂苷组上述损伤有一定程度的改善。结论山茱萸总皂苷可改善糖尿病大鼠睾丸功能,并在一定程度上减轻其病理损伤。
【Objective】The present study was to investigate effects of total saponins from cornus officinalis sieb.on the testis in STZ-induced experimental diabetic rats.【Methods】Male Sprague Dawley rat was i.p administered by streptozotocin（STZ） at a single dose of 60 mg/kg to induce diabetic mellitus.On the 28th day the animals with serum glucose level more than 16.7 mmol/L were selected and divided into three groups：diabetic group,CTS treated group（80 mg/kg,p.o）,methyltestosterone treated group（3 mg/kg,p.o）,12 rats each group.A non-diabetic group was set as a control.0.5% CMC-Na suspension was administered for consecutive 28 days and only medium was taken in control and diabetic group.The testis,epididymis,seminal vesicle of rats were weighed and the ratio of organ weight/ body weight was calculated.Glucose,testosterone in the serum and special enzymes such as lactate dehydrogenase（LDH）,acid phosphatase（ACP）,and γ-glutamyltranspeptidase（γ-GT） activity in the testis tissue were determined,as well as the testis block was stained by haematoxylin-eosin and observed under light microscope.【Results】Rats in diabetic group shown increased glucose level and declined testosterone in the serum,decreased LDH,ACP,γ-GT activity in the testis,compared with the control.Atrophied seminiferous tubules,decreased spermatogenic cells were shown in the testis of rats in diabetic group.These alterations were improved in total saponins treated group.【Conclusion】Total saponins of cornus officinalis sieb.could ameliorate the dysfunction of testis in diabetic rats,and at-
