摘要
目的 :建立一种适合影像学研究的家犬局灶性脑梗塞疾病模型。材料和方法 :10只健康成年家犬 ,手术组 7只 ,假手术对照组 3只。股动脉插管 ,DSA透视下将导管选择到一侧颈内动脉 ,注入聚氯乙烯酒精颗粒 (polyvinylalcohol,PVA)栓子。假手术对照组除不注入栓子外 ,其他操作步骤同手术组。分别从DSA、MRI、神经功能评分、形态学检查等方面评价模型效果。结果 :手术组动物术后出现不同程度的神经功能缺陷 ,MRI示同侧大脑中动脉区信号改变 ,DSA造影示血管中断。形态学检查出现相应缺血改变。假手术组动物未见异常改变。结论 :该种家犬脑梗塞疾病模型适于影像学及其他亚急性、慢性脑缺血实验研究。
Purpose:The experiment was to establish a new canine model of focal cerebral infarction, and be suited for the study of medical imaging especially. Materials and Methods:10 dogs were selected into the study, among them seven dogs were included in experiment group and another three belonged to sham-operation group.The femoral artery was punctured and a catheter was introduced and advanced under DSA fluoroscopy until the tip was in the cavity of internal carotid arteria. Cerebral infarction was produced by injection PVA particles. The sham-operation group underwent the same process except injection of embolus. The model was evaluated by DSA,MRI,neurological deficit score and light and electron microscope. Results: After operation,all experimental dogs showed neurological deficit ranging from mild hemiparesis to death and ischemic morphology changes were observed under light or electron microscope. High signal and decreased blood perfusion were observed in four dogs at MCA supplied area on MR imaging and the blood vessel in the area was seen discontinue on DSA. No ischemic lesions could be found in animals of sham-operation group. Conclusion:The canine model of cerebral ischemia is a reliable choice for medical imaging study and clinical follow-up study of subacute-chronic cerebral ischemic lesions.
