摘要
目的 :建立反相高效液相色谱法测定环孢素A (CsA)血药浓度。方法 :色谱柱 :Kromasil C18(5 μm ,4 6mm× 2 5 0mm) ,流动相 :乙腈 -水 (80∶2 0 ) ,流速 :1 4ml/min ,柱温 :70℃ ,波长 :2 15nm。结果 :全血最低检测浓度为 10ng/ml,CsA血药浓度在 4 6～ 14 72ng/ml范围内与峰面积有良好的线性关系 ,平均回收率 (95 16± 3 87) % ,日内CV≤ 6 8% ,日间CV≤9 6 % (n =5 )。结论 :方法简便、快速、准确 ,可用于CsA的药代动力学研究。
Objective: To determine the blood concentration of cyclosporin A (CsA) by establishing reversed high performance liquid chromatography (HPLC). Methods: Chromatography column: kromasil C18 (5μ, 4.6 mm×250 mm). Mobile phase: mixture of acetonitrile and water (80∶20). Flow rate: 1.4 ml/min. Column temperature: 70℃. Wavelength: 215 nm. Results: The lowest detected concentration of the whole blood was 10 ng/ml. CsA blood concentration had good linear relationship with the peak area from 46 ng/ml to 1472 ng/ml. The average recovery rate was 95.16±3.87%. The coefficient of variation (CV) within a day was ≤6.8% and between days was≤9.6% ( n =5). Conclusion: This method is easy, fast and accurate. It can be used in the pharmacokinetics study of CsA.
