摘要
目的 建立单步萃取反相高效液相色谱法测定人全血中环孢素A(CsA)浓度。方法 采用反相高效液相色谱法 ,罗通定为内标 ,YWG -C18色谱柱 ,乙腈 -甲醇 -水 -异丙醇 (30∶10∶6 0∶1.5 )为流动相 ,流速 1.0ml/min ,柱温 70℃ ,检测波长 2 10nm。结果 CsA全血浓度在 5 2 .6 0～ 10 5 2 .0 0ng·ml-1范围内线性关系良好 ,r =0 .9996 ,最低检测浓度为 2 0ng·ml-1,平均回收率96 .33% ,日内和日间RSD均低于 5 %。结论 本法简便灵敏、准确可靠、经济 。
Aim To establish a method of RP-HPLC for determination of cyclosporin A in human whole blood by single-step extraction.Methods A RP-HPLC was carried out on YWG-C 18(200 mm×4.6mm,10μm).Rotundine was used as the internal standard.The mobile phase was composed of acetonitrile-methyl alcohol-water-isopropyl alcohol(30∶10∶60∶1.5),the flow rate was 1.0ml/min,the temperatrue of column was set at 70℃,and the detecting wavelength was 210nm.Results The assay was shown to produce linear calibration curves over the range from 52.60 to 1 052.00ng·ml -1 for cyclosporin,r=0.999 6.The lowest plasma limitation was 20ng·ml -1.The average recovery was 96.33%,and RSD within a day and between days did not exceed 5%.Conclusion The method is found to be convenient,accurate and ecnomical for determination of cyclosporin A in human whole blood.
