摘要
目的 :采用RP HPLC法测定环孢素A血药浓度。方法 :色谱柱为HypersilBDSC18柱 ( 4 .6mm× 15 0mm ,5 μm) ,乙腈 水 ( 62∶3 8)为流动相 ,柱温 70℃ ,流速 1.8ml·min-1,检测波长 2 0 5nm。结果 :CsA血药浓度在 5 0~ 12 0 0ng·ml-1范围内 ,浓度与峰面积比有良好的线性关系 ,最低检测浓度为 10ng·ml-1。平均回收率为 ( 97.5± 5 .8) % (n =15 )。日内RSD≤6.8% ,日间RSD≤ 9.6%。结论 :方法简单、快速、准确 ,可用于环孢素的临床药动学研究。
OBJECTIVE To establish RP HPLC method to determine the whole blood concentration of cyclosporine A.METHODS A Hypersil BDS C 18 column was adopted and the temperature was set at 70℃. Mobile phase was a mixture of acetonitrile and water (62:38) and flow rate was 1.8 ml·min -1 . The UV detector was operated at 205 nm?RESULTS The assay procedure was shown to produce linear calibration curves over the range 50 ng·ml -1 to 1 200 ng·ml -1 in blood( r = 0.9996 ). Within the range, the recovery rate was ( 97.5 ± 5.8 )%( n =15). The limit of detection was 10 ng·ml -1 .The coefficients of variation within a day and between days did not exceed 6.8 %, 9.6 %,respectively.CONCLUSIONS This method is found to be easy, convenient and precise for clinical pharmacokinetic study of cyclosporine A.
