摘要
目的:通过体外培养的方式,观察长效β2鄄受体激动剂福莫妥罗(formoterol)、白三烯受体拮抗剂孟鲁斯特(montelukast)和扎鲁斯特(zafirlukast)3种药物对哮喘患者外周血单个核细胞(PMBC)分泌白介素鄄4(IL鄄4)、白介素鄄5(IL鄄5)的影响。方法:哮喘患者26例,正常对照组10名。分离受试对象之外周血单个核细胞并接种于细胞培养板,药物孔相应加入montelukast、zafirlukast或formoterol预培养2h后,所有培养孔加植物血凝素(PHA)刺激培养48h,收集上清液,用ELISA法测定培养上清液中IL鄄4、IL鄄5的浓度并进行相应的统计学处理。结果:①哮喘组PBMC单纯经PHA刺激后,平均IL鄄4、IL鄄5的分泌水平高于健康组。②Montelukast、zafirlukast可导致哮喘组IL鄄5分泌下降,且有统计学意义。结论:Montelukast和zafirlukast均能抑制IL-5的分泌,前者还可能对IL鄄4的分泌也具有抑制作用,因而有利于纠正哮喘患者Th1/Th2的不平衡。本研究未见formoterol对IL鄄4、IL鄄5的分泌有影响。
Objective:To observe the effect of formoterol,montelukast and zafirlukast on peripheral blood mononuclear cells'(PMBCs)activity in secreting IL-4and IL-5in patients with asthma.Methods:Twenty-six patients with asthma and10normal controls were enrolled in this study.PBMCs were separated and inoculated on cell culture board,then formoterol,montelukast and zafirlukast were added by turn.Sample cells were cultivated for48hours and PHA was added.Cell culture supernatant was collected for detecting the concentration of IL-4and IL-5by ELISA.Results:①The levels of IL-4and IL-5secreted by PBMCs in patients with asthma were higher than that in normal controls.②Montelukast and zafirlukast significantly decreased the level of IL-5.Conclusions:Montelukast and zafirlukast could suppress the activity of PMBC in secreting IL-5in patients with asthma,the former may also inhibit the secretion of IL-4and it is useful to improve the imbalance of Th 1 /Th 2 .However,formoterol hasn't show such effect in this study.
