哮喘患者外周血单个核细胞白细胞介素-13 mRNA表达的研究 被引量:1

摘要 目的探讨哮喘患者外周血单个核细胞(PBMCs)白细胞介素(IL)-13 mRNA的表达和糖皮质类固醇激素治疗对其的影响。方法采用逆转录聚合酶链反应(RT-PCR)法分别检测34例发作期哮喘患者(其中14例激素敏感型和10例激素抵抗型分别口服泼尼松治疗1周)、18例缓解期患者和30名健康对照者PBMCs IL-13 mRNA的表达。结果52例哮喘患者和30名健康对照者PBMCs IL-13 mRNA的表达量分别为16.72±1.12和8.25±1.15,两组比较差异有统计学意义(P<0.05)。发作期和缓解期哮喘患者PBMCs IL-13 mRNA的表达量分别为20.72±1.02和13.67±3.15,两组比较差异有统计学意义(P<0.05)。在34例发作期哮喘患者中,轻、中、重度患者PBMCs IL-13 mRNA表达量分别为16.35±2.38、19.12±2.16、23.57±2.43,相互间比较差异有统计学意义(P<0.05)。激素敏感型哮喘患者经泼尼松治疗1周后PBMCs IL-13 mRNA表达量(21.03±3.77)较治疗前(15.67±2.67)明显下降,差异有统计学意义(P<0.05);激素抵抗或依赖型哮喘患者经泼尼松治疗1周前后PBMCs IL-13mRNA表达量分别为20.35±3.58和19.57±3.34,差异无统计学意义(P>0.05)。结论PBMCs IL-13参与哮喘发生的病理过程,并调节糖皮质类固醇激素的作用,可作为判断哮喘患者病变严重程度的指标之一。 Objective To explore the relationship of interleukin 13 (IL-13)mRNA expression of peripheral blood mononuclear eells(PBMCs)with clinical feature in patients with asthma and the effect of glueocorticoid on it. Methods Thirty four cases of asthmatic patients with acute asthma attack (in which, 24 cases took prednisone orally for one week), 18 cases of asthmatic patients in remission stage, and 30 cases of healthy volunteers were enrolled. The PBMCs were obtained and the expression of IL-13 mRNA was detected by reverse transcription polymerase chain reaction(RT-PCR). Results The IL-13 mRNA level in patients with asthma and healthy volunteers were 16. 72 ± 1.12 and 8. 25 ± 1.15 respectively, and they had significant difference (P〈0.05). The IL-13 mRNA level in asthma attack group and asthma remission group were 20.72 ± 1.02 and 13.67 ± 3.15 respectively, and they had significant difference ( P 〈 0. 05). The IL-13 mRNA level in mild asthma group, moderate asthma group, and severe asthma group were 16.35±2.38, 19.12±2.16 and 23.57±2.43 respectively, and they had significant difference(P 〈 0. 05 ). After prednisone treatment, the IL-13 mRNA level of PBMCs decreased significantly in steroid-sensitive asthma group(P 〈 0. 05), while it didn't found in steroid-resistant asthma group. Conclusion The IL-13 may take part in the pathological and physiological process and regulate action of glucocorticoid in patients with asthmat. The IL-13 mRNA level of PBMCs may act as an index in judging asthma severity.
