摘要
目的 : 探讨黑米皮对 Apo E基因缺陷小鼠动脉粥样硬化斑块形成及血脂的影响。方法 : 45只 Apo E基因缺陷小鼠随机分为 3组 :阳性对照组、黑米皮组和白米皮组 ,同品系同龄正常 C5 7BL/ 6J小鼠作为阴性对照组。以 AIN-93 G为基础饲料 ,黑米皮组和白米皮组分别加入 5 %黑米皮和 5 %白米皮。饲养 1 6w,经小鼠眼眶静脉取血后处死动物 ,测定主动脉窦脂质斑块大小和血脂。结果 : 黑米皮组的实验小鼠主动脉窦脂质斑块面积明显低于阳性对照组和白米皮组 (P<0 .0 5 ) ;黑米皮组血清中总胆固醇和 LDL-C的含量明显低于阳性对照组和白米皮组 (P<0 .0 5 ) ,阳性对照组和白米皮组 LDL/ HDL比值明显高于黑米皮组 (P<0 .0 5 )。黑米皮组血清中抗 Ox-LDL抗体明显低于阳性对照组和白米皮组 (P<0 .0 5 )。结论 : 黑米皮具有明显抗 AS作用 ,其作用机制与改善脂质代谢。
Objective: To observe the effect of supplementation of black rice outer layer fraction on atherosclerotic plaque formation and blood lipids. Methods: C57BL/6J mice(Control group, n=15) were fed AIN 93G purified diet. Three groups of apoE deficient mice consumed one of the following diets: AIN 93G purified diet(Positive group, n=15); AIN 93G purified diet with 5 g/100 g black rice outer layer fraction(BRF group, n=15); AIN 93G purified diet with 5 g/100 g white rice outer layer fraction(WRF group, n=15). The animals were fed different diets for 16 w, then blood was determined for lipids. Results: There were no visible atherosclerotic plaques in the aortic sinus of the control group. But the plaques of various degrees were visible in other three groups. Plaques were much more severe in positive group and WRF group than in BRF group (P<0.01). The levels of serum TC and LDL C in BRF group were lower compared with positive group and WRF group(P<0.01). Although both BRF and WRF groups had higher levels of HDL C compared with positive group, the BRF group had lower LDL/HDL(P<0.01). Serum anti OxLDL antibody levels decreased significantly in the BRF group compared to WRF group and positive group(P<0.01). Conclusion: Black rice outer layer fraction can decrease atherosclerotic plaque formation and improve lipid profile in ApoE deficient mice.
