抗生素对大豆未成熟子叶愈伤形成和体细胞胚胎发生的影响 被引量:10

摘要 用 6个大豆品种进行了抗生素敏感性试验 ,结果表明 :在培养基中加入 5 0mg/L卡那霉素可显著地降低出愈率。大豆不同品种对卡那霉素的反应在出愈率上表现相似 ,而在体细胞胚胎发生率上存在差异。培养基中含有 40 0mg/L头孢霉素时出愈率显著降低 ,而体细胞胚胎发生率因品种不同而不同。用农杆菌侵染大豆未成熟子叶后在含有头孢霉素的培养基中培养时 ,体细胞胚胎发生率大幅度下降。本试验初步确定了较适合于大豆未成熟子叶抗性选择的卡那霉素浓度和头孢霉素的脱菌浓度。 The experiment of sensitivity on antibiotics was investigated with 6 genotypes of soybean. The results showed that frequency of callus fromation was decreased significantly when the medium was added with 50mg/L kanamycine. The response of soybean genotypes was similar in frequency of callus formation and was different in frequency of somatic embryogenesis. When medium contained 400mg/L cefotaxime, the frequencies of callus formation were dropped, however, the variation of somatic embryogenesis frequencies was different among different genotypes. The frequencies of somatic embryogenesis were declined obviously when immature cotyledon was cultured in medium containing cefotaxime after Agrobacterium-mediated. We provide an ideal selecting press of kanamycine and concentration for inhibiting Agrobacteriumof cefotaxime in immature cotyledon of soybean.
