应用抑制性消减杂交方法克隆人肝细胞癌差异表达基因 被引量:8

Cloning the genes differently expressed in human hepatocel-lular carcinoma by suppression subtractive hybridization

摘要 Diatchenko等[1]报道了一种新的快速克隆差异表达基因cDNA的方法,称为抑制性消减杂交(SSH),十分适用于克隆造成某种特殊细胞表型的特异表达基因,例如正常和病变之间的基因表达差异分析.人肝细胞癌(HCC)是我国高发的恶性肿瘤之一,其发生发展与多种基因的突变与表达异常有关.我们应用抑制性消减杂交方法克隆HCC与癌旁非癌组织之间差异表达基因的cDNA,为今后获得HCC疾病易感基因及其功能研究奠定实验基础.
