烧伤小鼠肝组织糖皮质激素受体的变化及安定-氯胺酮的影响 被引量:2

摘要 目的 研究烧伤小鼠 2 4h内肝组织糖皮质激素受体 (GR)时相变化特点 ,初步探讨安定 氯胺酮麻醉对创伤应激的调控作用。方法 将 80只BALB/C小鼠随机分为正常对照组、麻醉对照组、烧伤组、预处理组 (麻醉后 15min烧伤 )及后处理组 (烧伤后 15min麻醉 )。烧伤组给予背部Ⅲ°15 %~ 2 0 %烧伤 ,伤后 2 ,4,6 ,12 ,2 4h取血和肝脏 ;麻醉对照组、预处理组及后处理组均采用安定 氯胺酮麻醉 ,烧伤或麻醉后 4h取血和肝脏。测定血清皮质酮浓度 ,采用Westernblot在蛋白水平测定肝组织GR。结果 烧伤后小鼠血清皮质酮浓度明显升高 ,4h达到高峰 ;肝组织GR明显降低 ,6h降至最低。安定 氯胺酮麻醉明显抑制烧伤应激引起的血清皮质酮和肝组织GR的变化 ,甚至烧伤后麻醉 ,也有同样的作用。结论 烧伤应激引起小鼠血清皮质酮水平明显升高 ,其受体在蛋白水平显著降低。安定 氯胺酮麻醉对其具有抑制作用。 Objective To observe the change of glucocorticoid rceptor(GR)in liver and study the effect of diazepam-ketamine anesthesia in burned mice.Methods 80 BALB/c male mice were divided randomly into five groups:control,anesthesia control,burn,pretreatment and posttreatment groups.Burn group suffered 15%~20% thickness third degree burn injury.Anesthesia control group received diazepam-ketamine anesthesia only.Pretreatment group and posttreatment group received diazepam-ketamine anesthesia initiated 15 min before and after burn injury respectrely.During the 24h after burn,serum corticosterone concentration and hepatic GR were detected in burn group.At 4 h after burn or anesthesia,those were detected in anesthesia control,pretreatment and posttreatment groups. Results After burn injury,serum corticosterone level was markedly elevated at 4 h and hepatic GR was markedly downregulated at 6 h.The increased serum corticosterone and reduced GR responses to burn injury were markedly suppressed in the pretreatment and posttreatment groups.Conclusion Serum corticosterone level is markedly elevated and hepatic GR is markedly downregulated in burned mice.It is not principal machanism about downregulation of GR at mRNA level.Diazepam-ketamine anesthesia inhibits the response in mice with burn injury.
