摘要
目的 研究安定 氯胺酮复合麻醉对烧伤小鼠肝组织糖皮质激素受体的影响 ,初步探讨其对创伤应激的调节作用。方法 雄性BALB c小鼠 ,随机分为正常对照组、烧伤对照组、麻醉对照组、预处理组及后处理组。麻醉采用安定 氯胺酮复合麻醉 ,背部 15 %~ 2 0 %Ⅲ度烧伤。烧伤后 4h测定血清皮质酮浓度 ,用Westernblotting在蛋白质水平测定肝组织糖皮质激素受体。结果 烧伤对照组血清皮质酮浓度明显升高 ,肝组织糖皮质激素受体明显降低。预处理组与后处理组血清皮质酮浓度均低于烧伤对照组 ,肝组织糖皮质激素受体与正常对照组相差不明显 ,预处理组与后处理组相差不显著。结论 烧伤应激引起小鼠血清皮质酮明显升高 ,其受体在蛋白质水平明显降低。安定 氯胺酮复合麻醉对其具有一定的调节作用。
Objective To evaluate the effect of diazepam ketamine combined anesthesia on hepatic glucocorticoid receptor (GR) in burned mice. Methods BALB/c male mice were randomly divided into five groups: control; burn control, receiving 15%-20% thickness third degree burn injury; anesthesia control, receiving diazepam-ketamine combined anesthesia only; pretreatment, with diazepam ketamine combined anesthesia initiated 15 min before burn injury; and posttreatment, with diazepam ketamine combined anesthesia initiated 15 min after burn injury. At 4 h after burn injury or anesthesia, serum corticosterone concentration was examined and hepatic GR was detected by Western blotting. Results After burn injury, serum corticosterone level increased markedly whereas hepatic GR decreased markedly. The increased serum corticosterone and reduced GR responses to burn injury were markedly inhibited in the pretreatment group. The inhibitory effect was also found in the posttreatment. Conclusion Diazepam ketamine combined anesthesia is of regulating effect on the remarkable increase of serum corticosterone level and the remarkable decrease of glucocorticoid receptor at protein level in mice due to burn injury stress.
