严重烫伤大鼠肝组织核蛋白中糖皮质激素受体表达的变化及地塞米松对其的影响 被引量:11

摘要 目的 观察大鼠严重烫伤后早期肝组织核蛋白中糖皮质激素受体 (glucocorticoidre ceptor,GR)的表达变化 ,并探讨大剂量地塞米松对GR核转位的影响 ,为严重创伤时外源性糖皮质激素的合理应用提供可靠实验依据。 方法 结合Westernblot和免疫组织化学染色技术 ,检测大鼠严重烫伤后早期 4 8h内肝组织核蛋白中GR的表达变化情况 ;于烫伤后不同时间腹腔注射地塞米松 (5mg kg) ,通过Westernblot分析地塞米松使用前后肝脏GR由胞浆向胞核的转位改变。 结果 大鼠严重烫伤后 15min ,肝组织核蛋白中GR的表达就已开始降低 ,1h达最低值 ,显著低于正常对照 ,之后逐渐回升 ,12h恢复正常 ,2 4h后又明显高于正常对照 ;严重烫伤后 4~ 12h ,腹腔注射大剂量地塞米松可使肝脏组织核蛋白中GR的表达显著增多 ,而烫伤后 1h内和 2 4h后应用地塞米松对核内GR的表达无显著影响。 结论 严重烫伤可引起大鼠肝组织细胞核内GR的表达显著下降 ,在烫伤后 4~ 12h应用大剂量地塞米松可通过促进核转位来提高GR在胞核内的表达。 Objective To observe the expression changes of glucocorticoid receptor (GR) in the nucleuses of the liver, and to investigate the effect of dexamethasone on the transfer of the GR from cytoplasm to nucleuses in severe scalding rats in order to provide experimental bases for clinical treatment of the severe traumas by using properly the GR. Methods Western blot and immunohistochemistry were used to detect the expression of the GR in the nucleuses of the liver during 48 hours after scalding. The transfer of the GR to the nucleuses induced by intraperitioneal injection of dexamethasone (5 mg/kg) at different time points after scalding was determined by the Western blot. Results The expression of the GR in the nucleuses of the liver significantly decreased from 15 minutes after severe scalding, reached the lowest at the 1st hour (significantly lower than the control), resumed to normal at the 12th hour and was much higher than normal after 24 hours. The transfer of the GR to the nucleuses was evidently increased after intraperitioneal injection of high dose of dexamethasone during 4-12 hours after severe scalding. There was no effect of dexamethasone on the transfer of the GR within 1 hour and over 24 hours after scalding. Conclusions Severe scalding can significantly decrease the expression of the GR in the nucleuses of the liver. In 4-12 hours after scalding, the expression of the GR in the nucleuses can be improved by high dose of dexamethasone which may promote the transfer of the GR from cytoplasm to the nucleuses.
