摘要
目的 研究低氧对复合培养的平滑肌细胞中Janus激酶 (JAK)基因表达的作用。方法 鼠肺微血管内皮细胞与肺动脉平滑肌细胞复合培养 ,采用半定量RT PCR技术检测常氧组、低氧 2、6、12h组的JAK1、JAK2和JAK3基因的表达水平 ,并对PCR产物进行测序。结果 低氧复合培养 2h组肺动脉平滑肌细胞中JAK1、JAK2、JAK3mRNA表达水平升高 ,在低氧 6h组达最高 ,与正常组比较均有明显差别。低氧 12h组的表达量低于 6h组 ,但高于正常组。RT PCR产物直接测序证实扩增产物片断与Genebank中相应序列相同。结论 低氧增强复合培养的肺动脉平滑肌细胞中JAK基因的表达 ,而JAK基因在低氧所致的肺动脉收缩和低氧肺动脉高压的信号转导中可能发挥重要作用。
Objective To study the effect of hypoxia on the expression of Janus kinases (JAK) mRNA in co cultured cocuttured pulmonowy arterial smooth muscle cells Methods Rat pulmonary arterial smooth muscle cells (PASMC) were co cultured with rat lung microvascular endothelial cells, and then randomly divided into 5 groups: normal group (N), 2 h hypoxia (H2), 6 h hypoxia (H6), and 12 h hypoxia (H12) The expression levels of JAK1 mRNA, JAK2 mRNA and JAK3 mRNA in the PASMC were detected with semi quantitative RT PCR The products were sequenced with Sanger's double strand DNA sequencing Results The expression levels of JAK1 mRNA, JAK2 mRNA and JAK3 mRNA increased in the H2 PASMC, came to the highest in H6, decreased in H12, but still higher than that of N group They were also expressed in co cultured PASMC under normoxia The sequences of RT PCR products were consistent with those in Genbank Conclusion Hypoxia reinforces the expression of Janus kinases in co cultured PASMC, and their genes possibly play important roles in the signal transduction between pulmonary artery contraction and pulmonary hypertension
