摘要
本文对长期在无血清培养基内传代培养的亲本瘤和杂交瘤细胞的某些生物学特性进行了监测。结果显示,长期(三个月以上)在DMI传代的瘤细胞,贴壁能力明显减弱,个别瘤系完全呈悬浮型生长,但重新置回含血清常规培养基培养,贴壁能力大部分可恢复到原来状态:诱发BALB/c小鼠腹水和皮下实体瘤的能力保持不变;在HAT选择培养基中培养,瘤细胞的死亡时间在DMI和RPS15对照两种培养基内基本一致;4个亲本瘤系8-氮鸟嘌呤筛选结果与含血清培养的瘤细胞相似;经DMI长期传代的杂交瘤系分泌抗体的能力基本保持不变。
It has been developed in this laboratory that a serum-free medium DMI is adaptive for long-term culture, freezing and resurgence in liquid nitrogen, and cloning of hybridomas and parental myelomas as well as for cell fusion. In this report, it is described that the myeloma cells grown in DMI for more than three months maintained their biological characteristics such as induction of ascites and subcutaneous somatic tumor in BALB/c mice and tolerance toward 8-azaguanine, etc. However, the ability of the tumor cells in attaching to glass wall was decreased, in which some of the serum components may play an important role. The selecting assay of these cells in HAT medium showed that the death time in DMI was similar to that in conventional 15% newborn calf serum-supplemented medium(RPS15) The hybridoma cells adapted in DMI secrete monoclonal antibodies in quantities comparable to those produced in RPS15.
