摘要
我们研制了一种适用于瘤细胞长期传代培养、冷冻复苏、克隆化生长和细胞融合的无血清培养基(DMI)。在此基础上,本文进一步报告了亲本瘤和杂交瘤细胞在DMI内的生长特性。6株瘤细胞中,2株生长曲线的迟缓期稍延长,5株对数生长期历时较长,提示DMI尚缺少有益于细胞生长的、存在于小牛血清中的某种或某些因子;6株瘤细胞稳定期显著延长,提示细胞密度达高峰的维持时间较长,有助于分泌较多的单克隆抗体。SP_2/0、P_3U_1、983和13F_3在DMI内的倍增时间长于在含15%小牛血清培养基内的倍增时间,653和32B_5在两种培养基中的倍增时间相近。
It has been developed in laboratory a serum-free medium, designated as DMI (abbreviated from the Department of Malaria Immunology) , Which is adapted for long-term culture, repository in liquid nitrogen, and cloning growth of tumor cells as well as for cell fusion. In this report, the growth characteristics of hyb-ridoma and parental myeloma cells in DMI were described. Of 6 tumor cell lines, the lag phase of the growth curve of 2 lines wds protracted and the lag phase of 5 lines procrastinated,indicating that DMI may lack some useful growth factor(s) present in calf serum. The stationary phase of all tested tumor cell lines lasted for several days, which suggested that hybridomas were able to secrete larger amount of monoclonal antibody in DMI than in conventional medium with calf serum ( RPS15 ) . The double time ( Td ) of SP2/0, P3U1, 983, and 13F3 in DMI was longer than in RPS15; whereas,the Td of 653 and 32B5 was similar in these two media.
