摘要
本文在无血清培养条件下,对长期在DMI中传代培养的大鼠亲本瘤细胞系IR983F及小鼠亲本瘤细胞系P3U1和653对胆固醇的需求进行了动态观察。结果显示,胆固醇对这些亲本瘤系体外增殖是绝对必需的,其理想浓度为5—15μg/ml;10μg/ml量的人低密度脂蛋白可完全代替所需的胆固醇。与亲本瘤细胞系相比,长期在DMI中传代培养的杂交瘤细胞和SP2/0癌细胞对胆固醇的需要不十分严格。
Cholesterol, a major lipid component of plasma membrane, is thought to have profound effects on the structure and function of cells. Most animal tissues are capable of synthesizing cholesterol de novo from acetate; however, there have been relatively few demonstrations of mammalian cells in vitro expressing an absolute requirement for an exogenous source of cholesterol, In this paper, it was shownthat rat myeloma cells IR983F and mouse myeloma cells P3Ul and 653 which had been cultivated in serum-free medium for more than 6 months required cholesterol in vitro for growth in DMI. Optimal growth occurred in cholesterol concentration of 5 to 15 μg/ml. Moreover, it was demonstrated that the cholesterol could be replaced by human LDL in a concentration of 10 μg/ml.In contrast to parental myeloma cells, hybridoma cells derived from mouse myeloma cells which had been cultivated for more than 6 months in DMI did not absolutely require cholesterol, which indicates that cholesterol-deficient medium may be an effective alternative to HAT medium for selecting hybridomas.
