摘要
作者利用神经胶质瘤组织中提取的碱性蛋白(GBP)作为抗原免疫豚鼠制备免疫核糖核酸(iRNA),用白细胞粘附抑制试验(LAI)鉴定其活性;同时探讨建立MTT-LAI方法取代~3H-Leu LAI方法的可行性.结果表明:使用GBP作为抗原制备的GBP-iRNA有良好的生物活性;用MTT-LAI和~3H-Leu-LAI两种方法所得粘附抑制指数无显著差异,表明前者可取代后者作为iRNA活性鉴定的方法.
Immune RNA (iRNA) was prepared from the lymphoid tissue of guinea pigs immunized with glioma basic protein (GBP) extracted from pooled human glioma tissues, and its immunoactivity was verified by 3-(4,5-dimethyl-thiazol-2-yl) 2,5-diphenyl tetrazolium bromide-leucocyte adherence inhibition (MTT-LAI), a non-radioisotopic version of leucocyte adherence inhibition assay. The results showed that GBP-specific iRNA could mediate LAI in response to brain tumor antigens, such as glioma and meningioma antigens. 3H-Leu-LAI, currently used in cell-mediated immunity assay in vitm, could be replaced by MTT-LAI.
