摘要
目的:探讨绿茶调节摄食抑制因子-1(Nesfatin-1)和胃饥饿素(Ghrelin)表达对肥胖小鼠糖脂代谢及体重的影响。方法:将30只小鼠随机分为普通饲料组(空白对照组,10只)和高脂饲料组(20只)。高脂饲料组在造模12周后分为肥胖模型组和绿茶干预组,各10只。绿茶干预4周后处死小鼠,观察干预前后摄食量、体重变化;快速葡萄糖测定法测定血糖;酶联免疫吸附试验(ELISA)测定血清C肽、甘油三酯、游离脂肪酸、Nesfatin-1、Ghrelin水平;油红O染色法检测肝内脂质含量;免疫组化染色法检测肝及肠组织Nesfatin-1、Ghrelin蛋白表达;实时荧光定量PCR(RT-qPCR)检测下丘脑Nesfatin-1、Ghrelin mRNA表达。结果:经过4周干预,绿茶干预组摄食量和体重与空白对照组及肥胖模型组比较下降(均P<0.05)。肥胖模型组游离脂肪酸、甘油三酯、C肽水平及葡萄糖耐量实验(OGTT)曲线下面积(AUC)较空白对照组上升,而绿茶干预组较肥胖模型组下降(均P<0.05)。肥胖模型组血清Nesfatin-1、肝肠组织Nesfatin-1蛋白及下丘脑Nesfatin-1 mRNA表达较空白对照组下降,血清Ghrelin、肝肠组织Ghrelin蛋白、下丘脑Ghrelin mRNA表达及肝组织脂滴含量较空白对照组增加(均P<0.05)。绿茶干预组血清Nesfatin-1、肝肠组织Nesfatin-1蛋白及下丘脑Nesfatin-1 mRNA表达较肥胖模型组升高,血清Ghrelin、肝肠组织Ghrelin蛋白、下丘脑Ghrelin mRNA表达及肝组织脂滴含量较肥胖模型组下降(均P<0.05)。结论:绿茶可能通过上调Nesfatin-1和下调Ghrelin表达调节糖脂代谢,减少摄食量,进而有效降低肥胖小鼠体重。
Objective:To explore the effects of green tea on the expression of Nesfatin-1 and Ghrelin on glucose and lipid metabolism and body weight in obese mice.Methods:Thirty mice were randomly divided into normal diet group(blank control group,10 mice)and high-fat diet group(20 mice).After 12 weeks of modeling,the high-fat diet group was randomly divided into obesity model group and green tea intervention group,with 10 mice in each group.Four weeks after green tea intervention,the mice were sacrificed.Food intake and body weight changes before and after intervention were observed.Blood glucose was determined by the rapid glucose assay.Serum C-peptide,triglycerides,free fatty acids,Nesfatin-1 and Ghrelin levels were measured by ELISA.Oil Red O staining was used to detect lipid content in liver tissues.Immunohistochemical staining was used to detect the expression of Nesfatin-1 and Ghrelin proteins in liver and intestinal tissues.RT-qPCR was used to detect the expression of Nesfatin-1 and Ghrelin mRNA in the hypothalamus.Results:After 4 weeks of intervention,the food intake and body weight of the green tea intervention group decreased compared with the blank control group and the obesity model group(all P<0.05).The levels of free fatty acids,triglycerides,C-peptide,and the AUC of glucose tolerance test(OGTT)in the obesity model group were higher than those in the blank control group,while those in the green tea intervention group were lower than those in the obesity model group(all P<0.05).The expressions of serum Nesfatin-1,Nesfatin-1 protein in the liver and intestine tissues,and Nesfatin-1 mRNA in the hypothalamus in the obesity model group were lower than those in the blank control group,while the expressions of serum Ghrelin,Ghrelin protein in the liver and intestine tissues,and Ghrelin mRNA in the hypothalamus,and lipid droplet content in liver tissues were higher than those in the blank control group(all P<0.05).The expressions of serum Nesfatin-1,Nesfatin-1 protein in the liver and intestine tissues,Nesfatin-1 mRNA in the hypothalamus in the green tea intervention group were higher than those in the obesity model group,while the expressions of serum Ghrelin,Ghrelin protein in the liver and intestine tissues,Ghrelin mRNA in the hypothalamus,and lipid droplet content in liver tissues were lower than those in the obesity model group(all P<0.05).Conclusion:Green tea may regulate glucose and lipid metabolism by up-regulating Nesfatin-1 and down-regulating Ghrelin expression,reduce food intake,and thereby effectively lower the body weight of obese mice.
